Effect of Oxygen Tension on the Activity and Function of MG-y3 Human Osteosarcoma Cells

Abstract
This study is to investigate the effect of oxygen tension on the activity and function of MG-63 human osteosarcoma cells by measuring cell proliferation, cell differentiation, total protein synthesis, IL-1¥â, IL-6, and TNF-¥á. The MG-63 cells are incubated in the environment of 37¡É, 5% CO2, 100% humidity. The mixed gases were composed of 10% O2, 5% CO2, 85% N2 in hypoxic group or 90% O2, 5% CO2, 5% N2 in hyperoxic group and 5% CO2, 95% air for control. After incubation in 37¡É for 2, 4, 6 days, cell proliferation was determined by [3H]-thymidine incorporation, cell differentiation was assayed by alkaline phosphatase (ALP) activity and total protein synthesis was determined by sulforhodamine. And measurement of IL-1¥â, IL-6 and TNF-¥á was performed by enzymeimmunoassay. Results of this study show that in low oxygen tension, cellular proliferation increases, whereas the ALP activity and protein synthesis do not change. In contrast, in hyperoxic conditions cellular proliferation is suppressed with concomitant increases in ALP activity and decrease in protein synthesis. IL-6, TNF-¥á, and IL-1¥â were increased rapidly in both hypoxic and hyperoxic group in a time dependent manner.
